Date Golden Gate Assembly Confirmation Growing three colonies of each plate in 6mL LB broth for 6 hrs. Miniprep Plasmids and measure the concentration. Concentrations are very low. Cut the plasmids with SpeI and NotI, Not band were seen on the gel.
