Another round

I'm retaking the data for the resealable flow cell. This serves two purposes; 1. I'm not sure the data from the last time is reliable due to the flow cell breaking and 2. this is a modified design that may work better and I need to see if it does.

Update Andy Maloney 12:41, 28 January 2011 (EST): I should note that I do think the data is correct for the first 2 runs in the older data. It's just that I think there was evaporation going on in the 3rd and 4th sets due to the flow cell breaking. This test is to see if in fact the first 3 and possibly more washings will work.

0th washing

No problems here. Everything is working as it should.

1st washing

Andy Maloney 13:00, 28 January 2011 (EST): The first washing looks good. Actually, it looks spectacular.

2nd washing

Andy Maloney 13:35, 28 January 2011 (EST): The damn slip chipped again. I think the problem is using the vinyl. It's not "sticky" enough. I think I'll try plastic wrap after lunch.

Also, I'm seeing the microtubules start to have their plus ends go into solution more often. This is making me believe that the second washing is the last one that I can use in this assay.

Another flow cell

Andy Maloney 15:34, 28 January 2011 (EST): So I got some regular old cellophane from the grocery store. I sealed a flow cell and it looks spectacular. I think this is the way to go.

Andy Maloney 18:09, 28 January 2011 (EST): I was able to take data with the cellophane cell. We shall see how it turns out.
