Stock solutions
Yesterday and today I prepared stock solutions for heavy hydrogen PEM and deuterium depleted PEM. The procedure/recipe for both of these are below.
Heavy Hydrogen PEM
I am going to call this H-PEM to indicate that it is made up of heavy hydrogen water or, D2O. I didn't call it D-PEM for reasons below.
- Preparation of a 10x solution of H-PEM
- 800 mM PIPES
- 10 mM EGTA
- 10 mM MgCl2
- 1.3544 M NaOH
- pH 7.30 [[Steve Koch 00:12, 27 August 2011 (EDT): NOTE!!! I am thinking that this is a mistake and the buffer should have been made to 6.48 !!! See: http://friendfeed.com/anthonysalvagno/dbc577ae/preliminary-tobacco-seed-growth-results-those]
- Filtered through a 0.2 um filter and aliquoted in 2 mL screw top cryogenic vials in the 4C fridge.
Deuterium depleted PEM
This is called DD-PEM.
- 800 mM PIPES
- 10 mM EGTA
- 10 mM MgCl2
- 1.2265 M NaOH
- pH 6.89
- Filtered through a 0.2 um filter and aliquoted in 2 mL screw top cryogenic vials in the 4C fridge.
