McClean: Phosphate Buffered Saline

Materials

  • sodium chloride (NaCl)
  • potassium chloride (KCl)
  • sodium phosphate, dibasic (Na2HPO4)
  • potassium phosphate, monobasic (KH2PO4)

Protocol

To make 1L of a 10x solution:

  • Add together:
    • 80 g sodium chloride (NaCl)
    • 2 g potassium chloride (KCl)
    • EITHER 14.4 g of anhydrous sodium phophate, dibasic (Na2HPO4) OR 14.4g (anhydrous) x 268.07 g/mol (heptahydrate) / 141.96 g/mol (anhydrous) = 27.2 g of heptahydrate sodium phophate, dibasic
    • 2.4 g potassium phosphate, monobasic (KH2PO4)
  • Adjust pH to 7.4 with HCl if necessary.
  • Bring to 1 L with dH2O.
  • Autoclave
