Reaction Conditions
Incubate mixture at 37oC for 30 minutes and inactivate at 65oC for 15 minutes on the heat block.

Used SphI to digest pKn004.P1. Used 5 μl pKn004, 0.5 μl enzyme, 2 μl buffer, and 12.5 μl water. Incubated for 1 hour 15 minutes at 37 degrees and inactivated at 65 degrees for 15 minutes. Put in 4 degree over night.
