Restriction digest

  1. Determine the concentration of your DNA using the NanoDrop
  2. Use the ThermoFisher Calculator to determine which buffer to use for your reaction
  3. Determine the volumes needed to add 5 μg DNA to a 20 μL reaction using the information gathered above. Use dH2O to make up to 20 μL
  4. Incubate as described by ThermoFisher
  5. Either store your DNA at -20°C or continue to perform gel electrophoresis to separate your sample
