  • Ghaemmaghami et al. (2003 PMID 14562106) measured 8480/cell by quantitative western of entire proteome.
  • By FCS, the cytosolic Fus3 concentration was estimated to be 179 nM. Maeder et al. 2007 PMID 17952059
    • Given a cell volume of 29 fL (see Cell_volume), this corresponds to ~3,120 molecules per cell.
  • By FCS, the cytosolic Fus3 concentration was estimated to be 66 ± 17 nM in the absence of pheromone, and 127 ± 22 nM 2 hours after pheromone treatment. Slaughter et al. 2007 PMID 18077328
    • Given a cell volume of 29 fL (see Cell_volume), this corresponds to ~1,150 molecules per cell in the absence of pheromone.
  • Thomson et al. (in preparation) measured 20,413 molecules/cell by careful western blot.

Thomson et al. made careful measurements using purified Fus3 as a protein standard for their Western blots, and accounted for more sources of experimental error than Ghaemmaghami et al. However, there is no reason to suspect the 1,150-3,120 molecules per cell measured by Maeder and Slaughter may be erroneous. Additionally, fluorescence measurements by Thomson et al. suggested a much lower abundance of Fus3 than the ~20,000 molecules/cell that they measured by immunobloting, although there were not able to quantify Fus3 abundance, and it's not clear whether their immunoblotting or fluorescence measurements were erroneous. Thus, all we can say is that the Fus3 abundance likely lies between 1,150 and 20,413 molecules/cell.
