Testing of Homemade S30 Cell Extract

1. Set up 4 reactions with the following composition
   • S30 Cell extract 16.2μl
   • Reaction buffer 30μl
   • Pyruvate kinase 3.1μl
   • rNTPs 1μl
   • DNA/ddH20 4μl
   • ddH20 5.7μl
2. Left at 37°C incubator
   • 1. Cell extract reaction with pTet-GFP
   • 2. Cell extract reaction without pTet-GFP (negative control)
   • 3. Cell extract reaction with pT7-GFP
   • 4. Cell extract reaction without pT7-GFP (negative control)
