The Free Genes Project has several DNA synthesis projects. Here are breakdowns of each.
GLOBAL ASSUMPTIONS
- It takes 2 sequencing reactions to sequence an average gene (1 build well)
- A 96-well build to completion takes 120 minutes of active labor
- In each project, 1/12 of the constructs fail, requiring more builds + sequencing
- Assembly and transformation costs $2.50 in reagents. Sequencing costs $3.25 (per deal with Twist)
- An average plate contains 100kbp of sequence
- By this logic, each 100kbp costs $936 and 130 minutes of labor
- Labor is not factored into costs (but if it was, it would only raise the cost by $43.5)
- It takes 360 minutes to set up the bioinformatics pipeline to do any of these tasks.
