1% Agarose gel containing self-assembly reactions of both dome and plate and purified dome samples. All samples are assembled in 12.5mM MgCl2 over a 17 hour non-linear ramp. Lane 1: 1Kb ladder, Lane 2: M13 control, Lane 3: 21nM plate unpurified, Lane 4: 5nM dome unpurified, Lane 5: 5nM dome purified, Lane 6: 5nM dome purified, Lane 7: 5nM dome purified, Lane 8: 1nM dome unpurified, It is obvious that the purification using freeze n’ squeeze was a succes as the staple cloud and aggregations are gone after purification.
