WaaL ligase PCR
- Used Barnes buffer instead of Pfu Ultra Buffer
- PCR taken at 48'C, 50'C and 52'C with the negative at 48'C
- Successful PCR (at last!) at all 3 temperatures. We get the same unspecific band as we have been getting previously (perhaps the sterile water is not so sterile??).
- will run a restriction digest on the 52'C PCR product overnight, later today.
