LAB 5 WRITE-UP

PCR Reaction Report

In the lab dna was taken and using a pipette, placed into tubes. These tubes were then placed into the PCR. In the PCR the dna is heated to seperate the two strands of in order to complete transcription. The pre lab was helpful and made the lab more efficient. The first stop extracted the liquid and released it. While the second removes the plastic tip.

Fluorimeter Procedure

Smart Phone Camera Settings

  • Type of Smartphone: I-Phone 6
    • Flash:off
    • ISO setting:800
    • White Balance:Auto
    • Exposure:Highest
    • Saturation:Highest
    • Contrast:Lowest


Camera set-up
In order to set up the camera properly, one must first make sure the cradle is at least 4 centimeters away from the flourimeter. Make sure the camera is as close to eye level as possible to ensure the best look at the SYBR Green. Make sure the flash is off and all the prerequisites are met. Finally be sure to eliminate all white light from the flourimeter to ensure the best gathering of SYBR Green data.


  • Distance between the smart phone cradle and drop = 4.5 cm


Placing Samples onto the Fluorimeter

  1. Using gloves, pick a slide and place it smooth side down inside the flourimeter and adjust for height of the camera.
  2. Add 80 microliters of the SYBR Green to the slide making sure to place it in between the first and second row on the slide, align the drop with the Blue LED light.
  3. Press the shutter button on the camera and close the flap to ensure no excess white light interferes with the SYBR Green.
  4. To retrieve the sample, REMOVE THE BOX!! NOT THE SMARTPHONE OR FLOURIMETER!Then use the pipettor to remove the sample of SYBR Green from the slide.
