Targetron for msa knockout

  • Double digested vector, insert (msa and control retargeted)
  • Run gel
  • Gel purify (elute with 40ul for insert, 50ul for vector)
  • Ligate (one hour at room temperature)
  • Transformation in to Dh5, RN4220
  • Plate in amp (Dh5), erm (RN4220)
  • Incubate at 37 (Dh5), 32 (RN4220)
