Fragment Analysis Submission

  • Submit samples from 23 June 2011 PCR2 (condition indicated by below)
  • Submit samples from 24 June 2011 ( below)

  • Frag Submission Map:

  • FedEx Tracking# 875047691856

  • UAGC Submission# KXBJ

Evaporation test for Shipping

  • used 22 June 2011 PCR (failed)
  • sealed with flat-top 8-well strip caps, left on Team ConGen computer 23 June - 27 June

  • Plate Weight
- Before (23 June) After (27 June)
Trial 1 19.44g 19.22g
Trial 2 19.41g 19.24g
Trial 3 19.36g 19.26g
Average 19.40g 19.24g

Conclusion

  • 0.16 g loss of liquid via evaporation after 4 days at room temperature
  • [math]\displaystyle{ 0.16 g \bullet \tfrac{1 mL}{1 g} \bullet \tfrac{1000 \mu L}{1 mL} = 160 \mu L }[/math]
  • 32 rxn * 10µL - 40µL gel = 280µL sample in the plate; so >1/2 liquid is lost!
  • BUT: there's still at least ~ 3µL left in all wells, which is plenty for fragment analysis
  • BUT: when shipping, plate will be cold, and will only take 2 days
  • thus, sealing this way is a-okay!
