- 1. Synthesize two buffers to purify Nickel Hemoglobin previously synthesized
- 1. 10 mM Tris (1.21g) and 50 mM NaCl (2.922g) were added, along with 900 mL of distilled water
- 2. The pH was brought to 9.5 using 1M NaOH and 3 M HCl, and the remaining 100 mL of distilled water were added
- 3. 10 mM Tris (1.21g) and 1M NaCl (58.44g) were added with 900 mL of distilled water
- 4. The pH was brought to 9.5 using 1M NaOH, and the remaining 100 mL of distilled water were added
- 5. 10 mM Tris and 50 mM NaCl were added along with 3,975 mL of distilled water
- 6. The pH was brought to 9.5 using 3M HCl, and the remaining 25 mL of water were added
- 7. The resulting buffer was used as a dialysis solution, exchanging previously synthesized Ni Hb in pH 8.5 solution
- 8. The Ni Hb was placed in two dialysis bags and placed in the pH 9.5 solution in a cold environment, left overnight
