Objective

  • Synthesize gold nanoparticles from 1/31 stock solutions of HAuCl4 (2.9 mM), and BSA (17.7 μM).

Description

10 mL and 1 mL mixtures with the same molar ratios were created.


1 mL Mixtures:

Au/BSA Molar Ratio 70:

.042 mL HAuCl4 + .1 mL BSA + .85 mL H2O

Au/BSA Molar Ratio 166:

.1 mL HAuCl4 + .1 mL BSA + .8 mL H2O


10 mL Mixtures:

Au/BSA Molar Ratio 70:

.42 mL HAuCl4 + 1 mL BSA + 8.5 mL H2O

Au/BSA Molar Ratio 166:

1 mL HAuCl4 + 1 mL BSA + 8 mL H2O


1 mL Mixtures were placed in a heat block, and the 10 mL mixtures were placed in an oven. Both the oven and heat block were set at 80 degrees Celsius.

Data

Both pairs of mixtures were kept at a constant 80 degrees for approximately 2.5 hours. The 1 mL mixtures were in a heatblock, and the 10 mL mixtures were in an oven.

  • 1 mL mixtures:

Au/BSA molar ratio 70 yielded a purple solution (A)

Au/BSA molar ratio 166 yielded purple fibers in clear solution (B)



  • 10 mL mixtures:

Au/BSA molar ratio 70 yielded a purple solution (C)

Au/BSA molar ratio 166 yielded dark purple fibers in clear solution (D)


Notes

  • Mixture D had dark purple fibers, that stuck to the sides of the test tube. This could have been an indicator of contamination in the test tube.
  • These solutions will be used to test the binding of dye to lysine.
