Assay for a functional luciferase using a Moonlight luminometer


  • Today in the morning, I measured if there was any emission of light in the following cultures, guided by the kind help of Chris Wood Phd., and the willingness of Susana López, Phd. to lend her luminometer. The measurements are as follows:

  • The Control sample implies a tube containing citratum sodium medium prepared as described here.


  • The Medium sample contains LB as a negative control.


  • The Bacteria sample is DH5-α without transforming plasmids (which served as a bacterial negative control); the rest of the cultures were transformed with the corresponding plasmid:
  1. Click Beetle Green Luciferase
  2. Click Beetle Red Luciferase
  3. Photinus pyralis wild type luciferase
  4. Photinus pyralis mutated luciferase under J23102 promoter





  • The second graph excludes the functional click beetle green luciferase. Colors for each sample are:
  1. Control -> Purple
  2. Medium -> Blue
  3. Bacteria -> Pink
  4. ClickBeetleRed -> Black
  5. PPlucwt -> Green
  6. PPmluc -> Orange
  7. PPmlucJ23102 -> MISSING!!!
