  • YFP time course: repeat with other concentrations of dox


YFP/miR-luc time course
> Plate JDS33-47 cells at ~1x105 in 5x 12-well plates; use U2OS plain medium

  • Plates 1, 2: 0.1 ug/ml dox; 8 time points
  • Plates 3, 4: 0.01 ug/ml dox; 8 time points
  • Plate 5: no dox

> Set up three wells per time point for triplicate trials
> +Dox (1μg/mL) time points:

  1. Wed, 6/22/11, 1 pm (55 hrs.)
  2. Wed, 6/22/11, 8 pm (48 hrs.)
  3. Thurs, 6/23/11, 8 am (36 hrs.)
  4. Thurs, 6/23/11, 8 pm (24 hrs.)
  5. Fri, 6/24/11, 8 am (12 hrs.)
  6. Fri, 6/24/11, 11 am (9 hrs.)
  7. Fri, 6/24/11, 2 pm (6 hrs.)
  8. Fri, 6/24/11, 5 pm (3 hrs.)
  9. no dox

--> Harvest cells at 8 pm for Flow Cytometry
