Tamura Sequence

  • Prepared LB Agar Ampicillin Plates

1. Added 6.25g of Difco LB and 3.75g of Bacto Agar to 250mL DI Water. Autoclaved at 121 degree Celsius. Added 250 uL 1000x Ampicillin (100 mg/mL) at 55 degrees Celsius to media and swirled. Poured approximately 25mL per plate. Allowed to harden, inverted.

  • Plating glycerol stocks

1. Plated on 6 LB Agar Amp plates each of the Tokyo G-Block glycerol stocks using glass pasteur pipets. Incubated at 37 degrees Celsius.
