High Stability Plasmid Development Abstract Transformation of pSB1T3/B0015 ligation Transformation I performed the transformation of the three ligations of the pSB1T3 plasmid (which confers resistance to Tetracyclin Tc) and the part B0015 (which confers resistance to Kanamicin Km) at ratios 1:50, 1:20 and 1:10. I used 40μL of competent cells and 2μL of the ligation reaction. I used our protocol. I will check the plated bacte4ria tomorrow and asses their growth. Petri dishes containes 75μL Km and 75μL Tc, resulting in a final concentration of 0.5μg/mL.
