UA Biophysics: Autoclave Instructions

To get authorization to use it, please send an email to Lab manager.

Before beginning any work on the equipment, make sure it is in good condition. If something does not seem right, should be reported to the lab manager.

BEFORE STERILIZATION:

  1. Material to be sterilized must be clean
  2. Wrap metal material and pipettes in paper
  3. Place a sterilization indicator in each flask or inside each wrapped pack.
  4. Load items within the boundaries of the tray so that they do not touch the chamber walls.
  5. Make sure that all instruments remain apart during the sterilization process.
  6. Fill the water reservoir with dH2O. Never fill higher than the base of the safety valve holder into the chamber. Under no circumstance should water be filled above the safety valve holder.

STERILIZATION:

  1. Turn ON the green switch, located on the front panel.
  2. Turn the red needle on the pressure gauge counterclockwise to 0 psi.
  3. Set the knob to the FILL WATER position. The knob should be turned in a clockwise direction only!
  4. When the water reaches the mark at the front of the chamber, set the knob to the STE (sterilize) position.
  5. Shut the door, move the Door Closing Device into position, and tighten. Do not overtighten the door as this may result in damage to the gasket.
  6. When the sterilization cycle is complete, buzzer will sound
  7. If no drying is required:
    1. Turn the knob to the Exhaust / Dry position.
    2. When the white needle on the pressure gauge has reached 0 psi, open the door.
    3. Remove the material and turn the knob to the “0” position.
  8. If drying is required:
    1. Turn the knob promptly to the Exhaust / Dry position. Do not allow the pressure to drop below 10 psi. The sooner the valve is turned to Exh / Dry at the end of the sterilization cycle, the more effective and efficient will be the drying.
    2. When the white needle on the pressure gauge has reached 0 psi the door can be opened.
    3. Unscrew the Door as far as it will go, and open about ¾ of an inch.
    4. Leave the knob in the Exhaust / Dry position.
    5. When the drying cycle is complete, buzzer will sound and the light turn off
    6. Remove the material and turn the knob to the “0” position.
