Thermostable DNA ligase

Purpose

  • Thermostable DNA ligases can be added to a PCR reaction to ligate extended oligos. This is used for some mutagenesis methods.

Procurement

  • Examples for thermostable DNA ligases are Taq ligase NEB and the Ampligase Epicentre.

Use

  • If you want to ligate extended oligos in a PCR reaction it is important to have them phosphorilated at the 5'-end. Oligos will only be ligated when they are assembled on a compementary strand.
