Goal: identify transcription start sites of the entire genome
- Isolate total RNA
- Reverse transcribe the RNA using a 5' fluorescently labeled primer
- Isolate ssDNA
- Mix ssDNA into a standard sequencing reaction
- Sequence the mixed product
