Goal: identify transcription start sites of the entire genome

  • Isolate total RNA
  • Reverse transcribe the RNA using a 5' fluorescently labeled primer
  • Isolate ssDNA
  • Mix ssDNA into a standard sequencing reaction
  • Sequence the mixed product
