Large Autoclave (Room 435: by media prep room)
- Ask Lab Manager for instruction before using for first time!
- Do not open door during a run. Escaping Steam presents a burn risk!
- If there is an emergency, turn autoclave off and leave it to vent itself!
- Always use log book
- Turn autoclave on
- Select cycle
- Cycle 1: Media less than 1L
- Cycle 2: Media 1L or more
- Cycle 3: Glassware
- Cycle 4: Biological waste
- Put items into autoclave in Nalgene autoclavable trays
- Close door
- Wait for ready light to come on
- Press start button
- Do not open door until cycle is fully complete
- Open door a crack to and let stem vent
- Turn off autoclave when finished
Small Autoclave (Room 409 by RNA room)
- Ask Lab Manager for instruction before using for first time!
- Do not open door during a run. Escaping Steam presents a burn risk!
- If there is an emergency, turn autoclave off and leave it to vent itself!
- Make sure that the internal water level inside the autoclave chamber is even with top of metal platform.
- If it is not fill with dH2O until it is. This will ensure your samples get autoclaved, and not burnt.
- Make sure external reservoir is filled so that the water level is between the high and low mark
- If it is not fill with dH2O or empty so that it is
- Place samples to be autoclaved into the chamber using metal cages to stack multiple samples
- Shut door and turn wheel until door is firmly locked
- Place magnetic key into receptacle
- Turn autoclave on
- Adjust time and temperature
- Press the start button
- Wait till run is completely finished before opening door
- Do not open door if red light is flashing, or the pressure gauge is above zero
- Turn off autoclave when finished
