1. Transform pCP20 (p202 for me) - this is ampR and ts - grow at 30C
2. grow colony at 30, then streak out on LB only plate, plate at 42
3. pick colony, grow up and streak out on lb only, lb/amp, lb/markertrying to remove, plate at 30.
4. save clone if it shows correct phenotype
