August 25th, 2011 Visualized on the gel: 1. Ladder 2. digested plasmid backbone 3. digested VEGF original 4. digested VEGF amplified We got a clear band for the plasmid backbone and faint bands between 800 and 1000bp for both original and amplified VEGF. So, we cut out the bands and ligated them together overnight. We tried four different combinations: (ratio: volume of plasmid backbone to volume of digested construct) A. 1:9 backbone:original VEGF B. 1:9 backbone:amplified VEGF C. 3:7 backbone:original VEGF D. 3:7 backbone:amplified VEGF
