Today I performed another ligation. I used the 3:1 (in:bb) ratio plus 50ng backbone only control. After the ligation, I long-transformed using DH5αT cells. I also made a no plasmid (cells + water) negative control to plate. I added SOC, allowed to shake for 1 hour, then plated them.
