Procedure
- Add all 100 or 150uL of an MC1061 competent cell tube to 400mL LB
- Grow up overnight on 37*C shaker
- split into two centrifuge bottles and centrifuged at 4200rpm for 14 minutes
- pour off supernatant
- wash each pellet with 10mL artificial sea water (ASW)
- pour off ASW
- add 10mL ASW to each bottle and resuspend pellets
- centrifuge at 4200rpm for 14 minutes
- pour off supernatant
- re-suspend pellets
- combine the contents of the two bottles
- create 2uL aliquots
