SWTH Rhodopsin Amplified DNA

  • PCR done by Uma 11/8/2016, Will Richardson Samples + Dan Inserillo Samples
    • Band in the PCR -ve well indicates DNA contamination

  • PCR done by Uma 11/14/2016, Will Richardson Samples
    • Test for contamination.
    • First 4 samples run using old reagents used in previous PCR.
    • Second 4 samples, changed water, primer mix, and Qiagen Master Mix
    • Contamination persists indicated by band in the PCR -ve well

  • PCR done by Winnie 11/15/2016, Will Richardson Samples
    • Test for contamination
    • All new reagents used
    • Contamination persists indicated by band in the PCR -ve well

  • PCR done by Winnie and Uma 11/16/2016, Will Richardson Samples
    • Test for contamination
    • Well 2 and 3 represent the repeated comparison of old reagents vs new reagents PCR -ves only, keeping the Qiagen Master Mix the same
    • Well 5 - 9 represent PCR done with reagents from the Pre-PCR room and with the Pre-PCR room pipettes
    • PCR preparation in the lab (Pre-PCR bench) still indicate contamination, however PCR preparation in the Pre-PCR room shows no contamination
    • However, extract negative shows a band, so there is still the possibility of contamination during DNA extraction
