BSA Magnetic Particles Day 5

Create BSA. AuNPs and magnetic BSA AuNPS

  1. Synthesize BSA –AuNPs into test tube that will contain a total of 5mL
  2. Au solution that has a final concentration of 0.25mM
  3. BSA solution that has final concentrations that reflects the following Au:Protein ratios
  • 105
  • 110
  • 115
  • 120
  • 125
  • 130
  • 135
  • 140
  • 145
  • 150
  1. Water up to 5mL
  2. Put into incubator for 4 hours at 80 degrees Celsiuis
  3. Add 30 mM FeSO4*7H2O, 400 mM KOH, and 100 mM KNO3

Separately make BSA- AuNPs the same way as in steps 1-4 but with ratios of:

  • 125:1
  • 130:1
  • 135:1

Notes

Once the gold was added to the solution within the ratios of 105- 150, the solution immediately turned purple indicating that nanoparticles were formed. After about 10- 15 minutes, a green precipitate that was initially believed to be fibers were formed. This is due to the reduction of the gold by the FeSO4*7H2O.
