Goal

Create a GFP-based sensor to measure the forces involved in cell motility.

Project Outline

  1. Express GFPuv with a C-terminal sortase tag.
  2. Replace two of GFP's β-barrel loops with fibronectin's RGD and synergy (PPSRN) loops.
  3. Coat PEG/PMMA gels with expressed GFP-RGD.
  4. Determine a force/fluorescence curve.
  5. Measure cellular motility forces in vivo.
