Science Questions/ To-do list

  • In the abstract level, how could we live with imperfect efficiency? No matter writing mechanism we choose in

the end, the fraction of cells whose genome is actually rewritten will likely to be significantly less than a hundred percent.

  • Directed evolution/ protein chimera could be used for constructing the library of enzymes and target site on DNA. Take a look at: engineering ZFN projects by Dana Carroll (Utah)/ David J Segal (UC Davis) and intergrase directed evolution by Michele Carlos (Stanford)
  • Are there DNA binding proteins in nature for any specific DNA sequence?...Bioinformatics/structural question question.
  • On in vivo directed evolution, we want to have the sequence to mark for highly mutation site to be out site the highly mutation site itself.
