Objective

  • Begin protein expression by preparing stock solutions.
  • Perform gold nanoparticle(AuNP) synthesis using 30 μM Bovine Serum Albumin (BSA), 4.17 mM Chloroauric Acid (HAuCl4), and H2O.

Description

  • Protein Expression: Prepare four 1 L solutions with the following in each:

1. 10 g Tryptone, 5 g yeast extract, 2 g glucose/L.

2. Filled to 1 L with H2O.

3. Autoclaved.

  • AuNP Synthesis:

1. Tube 1: 0.5 mL BSA (30 μM), 0.5 mL HAuCl4 (4.17 mM), 9 mL H2O.

2. Left Tube 1 in oven set at 80 degrees celsius for 2 hours.

3. Tube 2: 0.5 mL BSA (30 μM), 1 mL HAuCl4 (4.17 mM), 8.5 mL H2O.

4. Left Tube 2 in oven set at 80 degrees celsius and removed every 30 minutes for 10 minutes over the course of 2 hours.

Data

  • Tube 1: Purple protein fibers in clear solution.
  • Molar ratio of Au/BSA for Tube 1 = 139.
  • Tube 2: Grey/ Dark Green protein fiber aggregate floating near surface of test tube.
  • Molar ratio of Au/BSA for Tube 2 = 278.
