Checking Sequences for another round of ligations R24L6.1 and R24L6.2 are both good (take 6.1, it's a perfect alignment with RFP, showed up in the library search). R23L6.1 and R23L6.2 are both good (take 6.1, it's also a perfect alignment with RFP, showed up in the library search). R18L5.1 and R18L5.2 don't seem to have the inserts, only RFP R17L6.1 and R17L6.2 are both good, both perfect alignments to all parts R17L5.1 and R17L5.2 don't have inserts, only RFP R16L6.1 and R16L6.2 are both good (take 6.2, it has a perfect alignment to RFP, showed up in the library search). R16L5.1 and R16L5.2 both don't have inserts, only RFP Bottom line: All of L5 ligations are re-ligated backbone and do not have inserts. Do not repeat ligations with L5 backbone. Have finished and completed: R24L6.1 R23L6.1 R17L6.1 R16L6.2 Need to re-do: R18, R22 (already re-tried R22, but should re-retry it again for safety, low efficiency on the transformations).
