Objectives

  • Prepare stock solutions of HAuCl4 and BSA in water.
  • Prepare solutions at various ratios of Au to BSA from the stock solutions.

Stock Solutions

  • Prepared 15 μM BSA stock solution in water.
  • Prepared 10 mM HAuCl4 stock solution in water.
  • Calculations:
    • 25 mL of 10 mM HAuCl4 stock solution requires the following:
  MW HAuCl4: 393.83 g/mol
  Mass HAuCl4:
        25 mL × (10-3 L/mL) × 10 mM × (10-3 mol/mmol) × 393.83 g/mol = 0.0985 g HAuCl4
  Actual mass HAuCl4: 0.0971 g HAuCl4
  Actual concentration: 9.862 mM
        0.0971g × (1/393.83 g/mol) × (1/(25 × 10-3 L)) × (103 mM/mol) = 9.862 mM HAuCl4 in H2O
    • 10 mL of 15 μM BSA stock solution requires the following:
  MW BSA: 66430 g/mol
  Mass BSA:
        10 mL × (10-3 L/mL) × 15μM × (10-6 mol/mmol) × 66430 g/mol = 0.00996 g BSA
  Actua mass: 0.0105 g BSA
  Actual concentration: 15.806 μM
        0.0105g × (1/66430 g/mol) × (1/(10-3 L)) × (106 μM/mol) = 15.806 μM

Notes

  • Solutions of Au/BSA were wrapped in foil to prevent them from photoeffects.
  • After heating, the oven was turned off and the solutions were left to cool in the oven until needed.
