Trying to handle as many things as possible!!! Today, I checked the gel of my restrictions and as I observed the correct size in a reasonable concentration, I ligated them using the following reaction for a total of 20μl: -H2O --------------------> 7μL -Buffer ligase ----------> 4μL -Vector (plasmid) -----> 2μL -Insert (luciferase) ---> 6μL -Ligase -----------------> 1μL I left the ligations at 16°C ON.
