Repeating the luciferase band purification I repeated what I did yesterday (restriction and electrophoresis). This time, I was able to purify a 414.4μg band of ~1600bp, which is consistent with the described 1653bp of luciferase. After following the protocol, I ended with a 50μl tube with purified luciferase ready for ligation. I ended by doing a restriction assay with ECO R1 and XBA1 of the double terminator BBa_B0015 (the one augusto had worked in), and left it at 37°C ON. Tomorrow, I will do the ligation.
