Day 2: Preparing plasmid stocks


  • Today, I cultivated 4 E. coli DH5α strains containing different plasmids in 4ml of liquid LB medium with antibiotic (according to their respective cassette). The plasmids (and their codes) are:


17. pSB1C3: This is a high copy plasmid for use in assembly of BioBrick® standard biological parts. It's length is 2047bp and has a chloramphenicol resistance.

18. pSB1T3: This is a high copy plasmid for use in assembly of BioBrick® standard biological parts. It's length is 2473bp and has a tetracycline resistance.

19. BBa_I51020: A BioBrick base vector designed to facilitate construction of new BioBrick vectors from BioBrick parts. It's length is 2438bp and has an ampicilin resistance. Can be used with other vectors.

30. pSB4A5: A BioBrick standard vector with low copy pSC101 replication origin (BBa_I50042) and ampicillin antibiotic resistance marker (BBa_P1002).


  • Strains were incubated at 37°C for 4-5 hrs.
  • Jorge and I extracted plasmid according to the Roche's kit protocol.
