September 25, 2013 Objective Run SDS-PAGE to observe the amount of hemoglobin leftover from yesterday. Description Gel Electrophoresis: Lane Assignments 100x Diluted Hemoglobin & Dye Pepsin 1 Pepsin 2 Pepsin 3 Pepsin 4 Pepstatin 1 Pepstatin 2 Pepstatin 3 Pepstatin 4 100x Diluted Hemoglobin & Dye Dr. Fox's sample 100x Diluted Hemoglobin & Pepsin JMC's 12. 100x Diluted Hemoglobin & Pepstatin UV-vis Sample Prep Combine 0.75mL 1M perchloric acid with 0.75mL hemoglobin&pepsin and 0.75mL hemoglobin*pepstatin samples that sat overnight Allow reaction to proceed for one hour (until 2:16pm) Centrifuge for 15 minutes Take supernatant, run absorbance measurements Data <br.> Cell numbers start at 1 on the left and go to 12 on the right.
