12/28/10 ✓ Cell growth assay: seed cultures for growth assay for 126-3, 154-2, 132-8, FTRx ✓ Culture: split all KAH U2OS lines 1:10 ✓ ChIP: 128-8, 129-4 DNA purification day 2 (etOH ppt overnight) Cell growth assay > To complement B-gal assay > KAH126-1 done already (images collected) Remaining lines: KAH126-3 KAH154-2 KAH132-8 FTRx > Plate 6.26x103 cells per well in 6-well plates --> Add 62.5 μL of 1x106/mL suspension to 10 mL plain medium to make 6.25x103/mL dilution --> Add 1 mL cell dilution to 3 mL plain medium in each well --> Duplicate samples: 0 dox, 0.5 μg/mL dox*, 1.0 μg/mL dox Note: *Trying less dox to lessen potential over-expression of neg. ctrls.
