Fluorescence Measurements

  • In order to determine if our gold nanoparticles (AuNPs) have any promise for electron applications such as FRET (flourescence resonance energy transfer), the fluorescence spectra of the amino acid (AA) stock solutions, a Bovine Serum Albumin (BSA), an acridine orange solution, a fluorescein solution, and solutions with combinations of these, were all determined using a fluorescence spectrometer.

Notes

The general procedure for today was as follows:

  • We prepared 50mL solutions of 1 μM of acridine orange by first creating 1mM solutions and then performing a 1:1000 dilution. Calculations are below:

Acridine Orange: (Molarity)*(Desired Volume of Solution)*(Molar Mass of Acridine Orange)=(0.001 mol/L)*(0.050 L)*(301.81 g/mol) = 0.01509 g

The resulting solution was then diluted by a factor of 1:1000. 0.05 mL of the solution was added to 49.95 mL of DI water.

Fluorescein: (Molarity)*(Desired Volume of Solution)*(Molar Mass of Fluorescein)=(0.001 mol/L)*(0.050 L)*(332.31 g/mol) = 0.0166 g

The resulting solution was then diluted by a factor of 1:1000. 0.05 mL of the solution was added to 49.95 mL of DI water.

  • The fluorescence spectrometer was then set to use an excitation wavelength of 308 nm and emission windows of 8 nm and a scan speed of 50 nm/min. The emission scan started at 10 nm over the selected excitation wavelength and scanned a range of 200 nm.
  • The fluorescence of the stock solutions were measured.
