1. Prepare culture dishes with Ampicilin antibiotic.

  • Instructions are as indicated on April 20th.


2. PCR with the minimum Blue Promoter and suffix primers to amplify plasmid 30 and ligate Blue Promoter to this plasmid.

  • Reactives needed per sample:
PCR with RTTH polymerase
Reaction 1
HPLC -> 10
Buffer 3.3x -> 6
Mg(Ac)2 -> 3
dNTP’s -> 2
Primer FWD -> 2.5
Primer REV -> 2.5
DNA -> 4
Total -> 30
Reaction 2
HPLC -> 10.5
Buffer 3.3x -> 9
RTTH -> 0.5
Total -> 20
