TAR
- XF capture construct is proving very difficult to assemble for some reason. PCR products look great, no errors in the primers, and done in parallel with two other constructs + controls, but no transformants on selective media after electroporating into commercial EC100's or homebrews, or EPI300's with induction. Will repeat the construction of the capture vector to make sure things are ok.
- 2.3, 3.4 Yeast captured fragments of ZB118 were electroporated into EPI300's and induced this time for 7hrs (3ml - 50ml LB + tet). Midiprep yielded 10ug of DNA, plenty to play with for mapping, etc.
- Additional ZB118 was grown ON in 5ml LB/KAN because someone spilled all of my DNA when I was out to dinner.
