TAR

  • Did a proper zymoprep on 5ml yeast culture in YPD and isolated plenty of DNA for transformation again. This DNA did not yield any colonies in homebrew EC100's again!! but EC300 Epicentre, commercials yielded dozens to play with on selection (tet).
  • Innoculated colonies in LB tet for ON growth and charaterization. DNA was again in very low yield (did not know that the oriV was inducible (traf) in these cells...will try again with induction to yield more DNA).
