Objective

Today we'll be synthesizing lysozyme-AuNPs with salt added before being heated, Image the tests run yesterday, and prepare diluted solutions for Citric Acid, MES, 2,2 bipyridine, NaCl, KCl, MgCl2, CaCl2 from 1M to 100 mM, 10 mM, 1 mM. We will run an experiment with various concentrations on the BSA 30:1 ratio of lysozyme-AuNPs, in order to determine the minimum concentration of this variables to cause fiber formation.

Procedure

For the fibers, we first made stock solutions as it can be seen in the following figure.

Then we added the necessary volumes in order to create the Solutions to get the fibers desired after heating.

After this, we created stock solutions of 100mM,10mM, and 1mM of Citric Acid, MES, 2,2 bipyridine, NaCl, KCl, MgCl2, CaCl2 by diluting the 1 Molar solutions of these 1M solutions. Then 0.5mL of each variable were added to the 30,55, and 110 ratios, in order to observe their effect on the fibers.

Meanwhile, we took images of all the tests run yesterday, and we observed that all salts form fibers or aggregates that disperse in solution but settle overtime.
