Objective

Today's objective is to try to control formation of purple fibers and purple solution. So using the same ratio as on the 29th of November we are going to obtain:

  • Purple fibers for the 70 ratio of Au/BSA (instead of purple solution)
  • Purple solution with the 166 ratio of Au/BSA (instead of purple fibers)

To promote fibers formation, we are going to use temperature effect by cooling the solution into the ice. On the contrary, to trigger purple solution formation we are going to destroy potentially formed fibers by shaking the solution.

Protocol

  • Solution A: mole ratio of Au/BSA equal to 70
  1. 0.042mL of HAuCl4 (2.9mM)
  2. 0.1mL of BSA (17.7µM)
  3. 0.85mL of water
  4. Place it in the heatblock (80°C) for 1 hour and then place it into the ice
  • Solution B: mole ratio of Au/BSA equal to 166
  1. 0.1mL of HAuCl4 (2.9mM)
  2. 0.1mL of BSA (17.7µM)
  3. 0.8mL of water
  4. Place it in the heatblock (80°C) for 1 hour and then place it into the ice

Results

We obtained in both cases purple solution which means that we have been able to inhibit fibers formation in the solution with the 166 ratio; however we have not been able to promote fibers formation in the solution with the 70 ratio.
