Objective

Today we are going to test our previous results; according to all our previous results the mole ratio of Au/BSA trigger purple fibers and purple solution formation. We are going to test two ratios (70 and 166); according to our previous results we are supposed to oberve fibers with the 166 ratio and we are supposed to obtain purple solution with the 70 ratio.

Protocol

  • BSA Stock Solution (17.7µM)
  1. 0.0117g BSA + 10mL water
  • Ratio Gold/BSA = 70 (solutions A and B)
  1. 0.042mL of HAuCl4 (2.9mM)
  2. 0.1mL of BSA (17.7µM)
  3. 0.85mL of water
  4. Solution B at 80°C for 2 hours (constant temperature)
  5. Solution A removed every 30minutes from heat block and left 10 minutes at room temperature (discontinuous temperature)
  • Ratio Gold/BSA = 166 (solutions D and C)
  1. 0.1mL of HAuCl4 (2.9mM)
  2. 0.1mL of BSA (17.7µM)
  3. 0.8mL of water
  4. Solution C at 80°C for 2 hours (constant temperature)
  5. Solution D removed every 30minutes from heat block and left 10 minutes at room temperature (discontinuous temperature)

Results

As a result we observed that:

  • Solution A: we obtained a purple solution (with a 70 ratio and discontinuous temperature)
  • Solution B: we obtained a purple solution (with a 70 ratio and constant temperature)
  • Solution C: we obtained a clear solution with purple aggregate (all fibers are sticked together) (with a 166 ratio and constant temperature)
  • Solution D: we obtained purple fibers spread through a clear solution (with a 166 ratio and discontinuous temperature)
