Objective

Today's objective is to get a stable purple solution at the end of the Gold NPs synthesis.

Protocol

  1. We prepared 4 tubes:
    1. Tube 1: 0.1ml of BSA + 0.1ml of HAuCl4 + 0.8ml of water
    2. Tube 2: 0.1ml of BSA + 0.1ml of HAuCl4 + 0.8ml of Tris buffer (pH 9.36)
    3. Tube 3: 0.1ml of BSA + 0.05ml of HAuCl4 + 0.85ml of water
    4. Tube 4: 0.1ml of BSA + 0.05ml of HAuCl4 + 0.85ml of Tris buffer (pH 9.36)
  2. We placed the 4 tubes in the heatblock during 2 hours at 80°C
  3. We turned off the heatblock and left the solution inside so that the temperature decreases slowly

Results

After 2 hours we observed that:

  1. Tube 1: ratio of BSA:gold of 1:1; we obtained several purple fibers
  2. Tube 2: ratio of BSA:gold of 1:1; nothing happened, the solution remained incolore
  3. Tube 3: ratio of BSA:gold of 2:1; we obtained a clear purple solution (same results as on the 28th of September)
  4. Tube 4: ratio of BSA:gold of 2:1; nothing happened, the solution remained incolore
