Objective

  • Today's objective is to create a labeled cysteine fluorophore. To do so, we're going to use the fluorophore 5-(Iodoacetamido)fluorescein (515.3 g/mol) and some cystein (121.16 g/mol).
  • We are also going to perform the four-point Probe. We're using group 1 results.

Protocol

We're going to use 5X more 5-(Iodoacetamido)fluorescein than cystein.

  1. Measure 1mg of 5-(Iodoacetamido)fluorescein (yellow powder)
  2. 1µL of cystein stock solution
  3. add buffer (Tris pH = 9.3) until fluorescein is solubilized (1ml)
  4. Vortex
  5. 100µL of DTT after 2hours of reaction

Data

The four-point probe technique didn't work well; we have not been able to determine the conductivity of the fibers.
