Methods

  1. Dilute thiol-modified TS primer with 1 mL of 10 mM tris buffer (pH 7.5).
  2. Add 1 μL of diluted thiol-modified TS primer to 5.6 mL of AuNP solution synthesized on 02/15/12.


Calculations:

ρ Au0 = 19.30 g/mL, diameter AuNP = 32.1 nm, MM Au0 = 196.967 g/mol


2 mL Au+3*(1 L/1000 mL)*(3.66 mmol/1 L)*(1 mol/1000 mmol) = 7.32*10-6 mol Au+3

V AuNP = (4/3)*π*(3.21*10-8 m/2)3 = 1.73*10-17 cm3

1.73*10-17 cm3*(19.30 g/1 mL)*(1 mol/196.967 g) = 1.695*10-18 mol Au/AuNP

7.32*10-6 mol Au+3/1.65*10-18 mol Au/AuNP = 4.32*1012 AuNP

4.32*1012 AuNP/20 mL solution = 2.159*1011 AuNP/mL solution

100 nmol DNA * (1 mol/109 nmol)*(6.02*1023 molecules/1 mol) = 6.02*1016 DNA

1 μL DNA * (1 ml/1000 μL)*(6.02*1016 DNA/1 mL)*(1 AuNP/50 DNA)*(1 mL/2.159*1011 AuNP) = 5.6 mL AuNP
